Laboratory centrifuges have several kinds of centrifugation

Laboratory centrifuge is mainly used for the separation of blood is known to all, virus research, the research of DNA, the purification of drugs, the solution density of different particle separation, radicalization immunity, biochemistry, agriculture and forestry sciences is the ideal equipment of production and scientific research units, but our laboratory centrifuge for different sample separation, centrifugal separation process each are not identical, The occasion is not the same. So what kind of common laboratory centrifuge centrifuge separation method?

① Differential centrifugal method

Differential centrifugal method is mainly to take the method of gradually increasing the centrifugation speed to separate the structure of different density in the sample, such as mitochondria, chloroplasts, proteins and so on!

Step 1: Set the starting centrifugation speed to be low and allow the larger particles to settle to the bottom of the tube while the smaller particles remain suspended in the supernatant.

Step 2: Collect precipitation, centrifuge the suspension at a higher centrifugal speed, settle the smaller particles, and so on, to achieve the purpose of separating particles of different sizes.

Advantages: large sample handling capacity, can be used for the initial separation of a large number of samples.

Disadvantages: when the separation of complex samples and the separation of high purity is required, the centrifugal times are many and the operation is complex. Because of the multiple cleaning, dissolution and re-precipitation of precipitation, it is easy to cause the intermediate loss, so the centrifugal resolution is poor.

② Gradient centrifugal method

Density gradient centrifugal method is to add the sample in the centrifugal tube in advance to increase the gradient medium for velocity sedimentation or sedimentation balance, under a certain centrifugal force to distribute the particles to some specific positions in the gradient, the formation of different zones of separation method.

Cesium chloride, sucrose and sucrose are commonly used in density gradient centrifugation.

Figure 3

Advantages: good resolution, good separation effect; The particles will not overstock deformation, can maintain the activity of the particles, and prevent the formed zone due to convection caused by mixing; It can be used to separate the particles with poor sedimentation coefficient and the particles with certain buoyancy density.

Disadvantages: sample concentration can not be too high; It takes a long time to reach equilibrium.

③ Analytical overspeed centrifugal method

The main purpose of analytical ultrafentrifugation is to study the sedimentation properties and structures of biomacromolecules rather than to collect a specific component. Therefore, it uses special rotors and detection means to continuously monitor the settling process of material in a centrifugal field. Analytical overspeed centrifuge mainly consists of an elliptical rotor, a vacuum system and an optical system. The rotor is connected to a high-speed drive by a flexible shaft that enables the rotor to form its own shaft as it rotates.

Advantages: accelerate the sedimentation rate of the particles in the liquid, and separate the matter with different sedimentation coefficient and buoyancy density in the sample.

Disadvantages: need to use the centrifuge to generate strong centrifugal force, to force these particles to overcome the diffusion of settling motion.

conclusion

The above three centrifugal separation is a more common laboratory separation method, are used in laboratory centrifuges. According to different samples and applications, they can be divided into industrial laboratory centrifuges and medical laboratory centrifuges.
